相分離液滴を基盤とするバイオマテリアル・デバイス開発

液-液相分離により形成される濃厚な高分子液滴は、高分子科学では古くから研究されてきた高分子に特徴的な状態です。近年では、細胞内で核酸やタンパク質などの生体高分子が高濃度に存在する一種のオルガネラとして注目を集め、生命科学やバイオマテリアル分野で活発に研究されています。本講演では、このような相分離液滴を基盤としたバイオマテリアル応用やデバイス開発について、最新の研究・開発動向を企業やアカデミアの研究者の方々からご紹介いただきます。

蛋白質の計算設計や超微量活性計測、化学修飾と状態制御などの急速な技術発展にともない、非天然の機能や天然を超越した活性を持つ蛋白質の開発が目覚ましい。そこで本ワークショップでは、新機能と超越活性を持つ蛋白質開発と応用に焦点を当て、最前線の研究を紹介する。
Rapid technological advances in computational protein design, ultra-trace activity measurement, and chemical modification have enabled remarkable progress in developing proteins with non-natural functions and activities that surpass those found in nature. This workshop focuses on the development and application of proteins possessing novel functions and transcendent activity, presenting cutting-edge research in this field.

人工蛋白質の計算設計や、クライオ電子顕微鏡を用いた蛋白質の構造解析など、ポリペプチドの構造の設計・解析技術が近年目覚ましく発展しています。一方、蛋白質の合成や機能設計のためには、フォールディングや分子運動などの速度論的特性を理解し、分子運動の加速や遅延などを制御することが重要です。ポリペプチドの超分子化学、医薬品・材料科学を今後発展させる上で、構造解析と速度論制御を結ぶ学際的な新領域の創出が重要であり、生体関連化学と、計算科学、構造解析技術、運動解析技術の分野で最先端研究に取り組む研究者により、各分野の最新の知見について共有し、今後の展望を議論するシンポジウムを開催します。

The biological properties of proteins largely rely on the higher-order structures. Correct folding of the unstructured polypeptide chains to the native structures allows for physiological functions, whereas misfolding-induced insoluble assemblies, aggregates such as amyloid fibrils, often cause a number of degenerative diseases. The process of proteins to form dense assemblies with liquid characteristics, namely liquid-liquid phase separation (LLPS), plays key roles in cellular functions such as signaling and gene expression. Understanding of the folding and assembling mechanisms is one of the central subjects in biological science, which is also helpful for drug design to combat pathogenic assemblies and recover abnormal protein functions. In light of bioengineering, the control of the assembling nature of proteins provides materials design based on (poly)peptides with bio-functionalities. Non-toxic fibrillar assemblies consisting of collagen helices and β-sheet stackings have been extensively developed to mimic extracellular matrices for tissue engineering and drug delivery. In addition, coacervates made of polypeptides have also gained attention as dynamic media with stimuli responses. Efficient combination of various chemical methodologies will give us deeper understanding of the polypeptide folding and assembly, and their controlling to advance the materials design and therapeutic applications. In this symposium, we highlight the recent progress in the chemical biology of protein folding, aggregation, and LLPS, and discuss chemical approaches to develop the biofunctional and therapeutic materials.

学術変革領域研究(B)「遅延制御超分子化学」は、分子からマクロスケールまで、自然界で広く見られる「遅延によって制御される機能と高次構造」から着想し、速度論制御を利用して機能的な非対称構造を人工構築する超分子化学の、基礎学理を構築することを目的とする研究領域です。分子スケールの非対称構造を有するタンパク質の構造構築原理を明らかにする生物物理学研究、細胞・オルガネラスケールの非対称構造の構築原理と機能を明らかにする分子生物学研究、生命に見られる非対称構造構築原理を物質合成と機能創出に導く材料科学研究を行ってきました。本企画では、本領域研究の成果を報告させていただくとともに、「遅延」から創られる生命機能と高次構造について化学の視点から広く討論し、「遅延制御超分子化学」のさらなる可能性と展望について議論する機会としたいと思います。

蛋白質の機能発現には、その構造に加え動きが密接に関わっている。一分子コンフォメーション変化や動的多量体・相分離形成など、蛋白質が見せる多様な分子スケールの動きを捉え、理解し、制御することが、化学を基盤とする生命科学、生体材料科学の新しい潮流を切り拓く。本企画では、溶液中や細胞内での蛋白質ダイナミクスを高い時間空間解像度で可視化し、その機能発現機序を理解する化学、計算科学、さらには生体内環境での蛋白質ダイナミクスを制御する分子材料科学、分子生命科学について、第一線の研究者から最新の研究を講演して頂く。蛋白質一分子の動き、さらには集合体の秩序的な連動や遅延現象に着目した生命機能の理解、生体模倣材料の開発につながる討論を行う。

タンパク質は熱力学的に最安定構造を取り機能発現するが、速度論的にトラップされた中間体構造が時折ミスフォールディングやタンパク質凝集体を惹起する。こういったタンパク質凝集体の蓄積は神経変性疾患をはじめ様々なミスフォールディング病を引き起こす。本講演ではフォールディング異常が引き起こす脳機能障害について、分子から個体までシームレスに議論することを目指し、最新の知見について紹介する。
Protein folding is intrinsically error-prone, and random coil polypeptides must be navigated via kinetically-trapped folding trajectories within the energy landscape to prevent aggregation. The accumulation of such aggregation causes various misfolding diseases including neurodegenerative disorders. In this workshop, we will discuss the latest findings with the brain dysfunction caused by aberrant proteins from a multidisciplinary perspective.

In recent years, brain science has made remarkable progress. Understanding neural circuits and elucidation of signal transduction processes at the molecular level are being carried out. Not only neuroscience but also mechanistic biochemical studies on neural diseases are progressing. Neurodegenerative diseases are one representative example, and the structure and dynamics of the causative protein are being elucidated at the single-molecule level. Integrating discussions between biophysical neuroscience and chemical research of the brain should address important unexplored issues such as the precise elucidation of brain function and the development of neuronal tissue regeneration technology.
